Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000005.10:g.33985072_33985075del, citing Invitae Variant Classification Sherloc (09022015): This variant occurs in a non-coding region of the SLC45A2 gene. It does not change the encoded amino acid sequence of the SLC45A2 protein. This variant is present in population databases (no rsID available, gnomAD 0.7%), and has an allele count higher than expected for a pathogenic variant. This variant has been observed in individual(s) with clinical features of oculocutaneous albinism type 4 (PMID: 30019506, 30809845, 32686196, 32969595; internal data). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. It has also been observed to segregate with disease in related individuals. Studies have shown that this variant alters SLC45A2 gene expression (PMID: 30019506). For these reasons, this variant has been classified as Pathogenic.
