NM_000051.4(ATM):c.308A>G (p.Tyr103Cys) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Y103C variant (also known as c.308A>G), located in coding exon 3 of the ATM gene, results from an A to G substitution at nucleotide position 308. The tyrosine at codon 103 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
