Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004360.5(CDH1):c.1916C>T (p.Thr639Ile), citing Ambry Variant Classification Scheme 2023: The p.T639I variant (also known as c.1916C>T), located in coding exon 12 of the CDH1 gene, results from a C to T substitution at nucleotide position 1916. The threonine at codon 639 is replaced by isoleucine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
