Uncertain significance for Brody myopathy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004320.6(ATP2A1):c.2045C>T (p.Ser682Leu), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces serine, which is neutral and polar, with leucine, which is neutral and non-polar, at codon 682 of the ATP2A1 protein (p.Ser682Leu). This variant is present in population databases (rs760340608, gnomAD 0.006%). This variant has not been reported in the literature in individuals affected with ATP2A1-related conditions. ClinVar contains an entry for this variant (Variation ID: 1007355). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant¬†is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
