Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 248812 control chromosomes (gnomAD). c.1874A>G has been reported in the literature in individuals (including a homozygous individual) affected with mucopolysaccharidosis Type 1 (example: Fernndez-Marmiesse_2014, Pineda_2014, Thomas_2021). These data indicate that the variant is likely to be associated with disease. Enzymatic activity measured from a homozygous individual show 2% alpha-iduronidase activity (Thomas_2021).
