Uncertain significance for Propionic acidemia — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000282.4(PCCA):c.1933G>A (p.Glu645Lys), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamic acid with lysine at codon 645 of the PCCA protein (p.Glu645Lys). The glutamic acid residue is highly conserved and there is a small physicochemical difference between glutamic acid and lysine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with PCCA-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0".
