Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_017654.4(SAMD9):c.334C>G (p.Gln112Glu), citing Ambry Variant Classification Scheme 2023: The p.Q112E variant (also known as c.334C>G), located in coding exon 1 of the SAMD9 gene, results from a C to G substitution at nucleotide position 334. The glutamine at codon 112 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
