Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_018474.6(KIZ):c.1102G>A (p.Glu368Lys), citing Invitae Variant Classification Sherloc (09022015): In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant has not been reported in the literature in individuals with KIZ-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change replaces glutamic acid with lysine at codon 368 of the KIZ protein (p.Glu368Lys). The glutamic acid residue is highly conserved and there is a small physicochemical difference between glutamic acid and lysine. Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown.
