Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_021167.5(GATAD1):c.231GGGCGG[3] (p.Gly80_Gly81dup), citing Ambry Variant Classification Scheme 2023: The c.237_242dupGGGCGG variant (also known as p.G80_G81dup), located in coding exon 1 of the GATAD1 gene, results from an in-frame duplication of GGGCGG at nucleotide positions 237 to 242. This results in the duplication of 2 extra residues (GG) between codons 80 and 81. This amino acid positions are highly conserved in available vertebrate species. In addition, this variant is predicted to be neutral by in silico analysis (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). The evidence for this gene-disease relationship is limited; therefore, the clinical significance of this alteration is unclear.
