Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_018474.6(KIZ):c.1196G>A (p.Gly399Asp), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glycine with aspartic acid at codon 399 of the KIZ protein (p.Gly399Asp). The glycine residue is moderately conserved and there is a moderate physicochemical difference between glycine and aspartic acid. This variant is present in population databases (rs767137895, ExAC 0.01%). This variant has not been reported in the literature in individuals with KIZ-related conditions. Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
