Uncertain significance for Developmental and epileptic encephalopathy, 1; Autosomal dominant nonsyndromic hearing loss 65 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001199107.2(TBC1D24):c.592A>T (p.Met198Leu), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces methionine with leucine at codon 198 of the TBC1D24 protein (p.Met198Leu). The methionine residue is moderately conserved and there is a small physicochemical difference between methionine and leucine. This variant is present in population databases (rs779920318, ExAC 0.02%). This variant has not been reported in the literature in individuals with TBC1D24-related conditions. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt TBC1D24 protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
