Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_181486.4(TBX5):c.100G>T (p.Ala34Ser), citing Ambry Variant Classification Scheme 2023: The p.A34S variant (also known as c.100G>T), located in coding exon 1 of the TBX5 gene, results from a G to T substitution at nucleotide position 100. The alanine at codon 34 is replaced by serine, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species, and serine is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
