Uncertain significance for Tall stature-scoliosis-macrodactyly of the great toes syndrome; Acromesomelic dysplasia 1, Maroteaux type — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003995.4(NPR2):c.749A>G (p.Tyr250Cys), citing Invitae Variant Classification Sherloc (09022015): ClinVar contains an entry for this variant (Variation ID: 1004546). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This missense change has been observed in individual(s) with autosomal recessive acromesomelic dysplasia (Invitae). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. This sequence change replaces tyrosine, which is neutral and polar, with cysteine, which is neutral and slightly polar, at codon 250 of the NPR2 protein (p.Tyr250Cys). This variant is present in population databases (no rsID available, gnomAD 0.003%).
