Uncertain significance for Gorlin syndrome; Medulloblastoma — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_016169.4(SUFU):c.1246G>T (p.Ala416Ser), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the SUFU gene (transcript NM_016169.4) at coding-DNA position 1246, where G is replaced by T; at the protein level this means replaces alanine at residue 416 with serine — a missense variant. Submitter rationale: This variant has not been reported in the literature in individuals with SUFU-related conditions. This sequence change replaces alanine with serine at codon 416 of the SUFU protein (p.Ala416Ser). The alanine residue is highly conserved and there is a moderate physicochemical difference between alanine and serine. This variant is not present in population databases (ExAC no frequency). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
