Uncertain significance for Fructose-biphosphatase deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000507.4(FBP1):c.799G>A (p.Ala267Thr), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces alanine with threonine at codon 267 of the FBP1 protein (p.Ala267Thr). The alanine residue is moderately conserved and there is a small physicochemical difference between alanine and threonine. This variant is present in population databases (rs576352790, ExAC 0.01%). This variant has not been reported in the literature in individuals affected with FBP1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
