Uncertain significance for Neutral lipid storage myopathy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_020376.4(PNPLA2):c.1266G>C (p.Trp422Cys), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces tryptophan with cysteine at codon 422 of the PNPLA2 protein (p.Trp422Cys). The tryptophan residue is highly conserved and there is a large physicochemical difference between tryptophan and cysteine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with PNPLA2-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0").
