Uncertain significance for Cerebral folate transport deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_016729.3(FOLR1):c.415T>C (p.Cys139Arg), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces cysteine with arginine at codon 139 of the FOLR1 protein (p.Cys139Arg). The cysteine residue is highly conserved and there is a large physicochemical difference between cysteine and arginine. This variant is not present in population databases (ExAC no frequency). This missense change has been observed in individual(s) with clinical features of FOLR1-related conditions (Invitae). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0").
