This variant lies in the RYR1 gene (transcript NM_000540.3) at coding-DNA position 9683, where C is replaced by T; at the protein level this means replaces alanine at residue 3228 with valine — a missense variant. Submitter rationale: This sequence change replaces alanine with valine at codon 3228 of the RYR1 protein (p.Ala3228Val). The alanine residue is highly conserved and there is a small physicochemical difference between alanine and valine. This variant is not present in population databases (ExAC no frequency). This missense change has been observed in individual(s) with clinical features of congenital myopathy (Invitae). In at least one individual the data is consistent with the variant being in trans (on the opposite chromosome) from a pathogenic variant.
