This variant lies in the SCN4A gene (transcript NM_000334.4) at coding-DNA position 4168, where A is replaced by G; at the protein level this means replaces methionine at residue 1390 with valine — a missense variant. Submitter rationale: This sequence change replaces methionine with valine at codon 1390 of the SCN4A protein (p.Met1390Val). The methionine residue is highly conserved and there is a small physicochemical difference between methionine and valine. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
