Submitter rationale: Variant summary: VWF c.4247T>A (p.Ile1416Asn) results in a non-conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant was absent in 251152 control chromosomes. c.4247T>A has been observed in individual(s) affected with dominant Von Willebrand Disease. These data indicate that the variant is likely to be associated with disease. A different variant affecting the same codon has been classified as pathogenic by our lab (c.4247T>C, p.Ile1416Thr), supporting the critical relevance of codon 1416 to VWF protein function.
