Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006231.4(POLE):c.3173A>T (p.Lys1058Met), citing Ambry Variant Classification Scheme 2023: The p.K1058M variant (also known as c.3173A>T), located in coding exon 26 of the POLE gene, results from an A to T substitution at nucleotide position 3173. The lysine at codon 1058 is replaced by methionine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
