Uncertain significance for Epidermolysis bullosa simplex 3, localized or generalized intermediate, with BP230 deficiency; Hereditary sensory and autonomic neuropathy type 6 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001374736.1(DST):c.19108C>G (p.Leu6370Val), citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site, but this prediction has not been confirmed by published transcriptional studies. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This sequence change replaces leucine with valine at codon 3747 of the DST protein (p.Leu3747Val). The leucine residue is moderately conserved and there is a small physicochemical difference between the two amino acids. The DST gene has multiple clinically relevant transcripts. This variant occurs in alternate transcript NM_015548.4, and corresponds to NM_001723.5:c.*106857C>G in the primary transcript. This variant has not been reported in the literature in individuals with DST-related conditions. This variant is not present in population databases (ExAC no frequency).
