Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_003001.5(SDHC):c.34C>G (p.His12Asp), citing Ambry Variant Classification Scheme 2023: The p.H12D variant (also known as c.34C>G), located in coding exon 2 of the SDHC gene, results from a C to G substitution at nucleotide position 34. The histidine at codon 12 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
