This variant lies in the BRCA2 gene (transcript NM_000059.4) at coding-DNA position 8345, where G is replaced by C; at the protein level this means replaces serine at residue 2782 with threonine — a missense variant. Submitter rationale: This sequence change replaces serine with threonine at codon 2782 of the BRCA2 protein (p.Ser2782Thr). The serine residue is highly conserved and there is a small physicochemical difference between serine and threonine. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
