NM_002528.7(NTHL1):c.20G>A (p.Arg7Lys) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R15K variant (also known as c.44G>A), located in coding exon 1 of the NTHL1 gene, results from a G to A substitution at nucleotide position 44. The arginine at codon 15 is replaced by lysine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
