Submitter rationale: This sequence change replaces glutamic acid, which is acidic and polar, with lysine, which is basic and polar, at codon 541 of the KCNT1 protein (p.Glu541Lys). This variant is present in population databases (rs753040489, gnomAD 0.001%). This variant has not been reported in the literature in individuals affected with KCNT1-related conditions. This missense change has been observed in at least one individual who was not affected with KCNT1-related conditions (internal data).
