Uncertain significance for Charcot-Marie-Tooth disease type 4 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_024577.4(SH3TC2):c.227G>C (p.Arg76Thr), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces arginine with threonine at codon 76 of the SH3TC2 protein (p.Arg76Thr). The arginine residue is highly conserved and there is a moderate physicochemical difference between arginine and threonine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with SH3TC2-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
