Uncertain significance for Spermatogenic failure 18; Ciliary dyskinesia, primary, 37 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_015512.5(DNAH1):c.9575C>T (p.Thr3192Met), citing Invitae Variant Classification Sherloc (09022015): This variant is not present in population databases (ExAC no frequency). This sequence change replaces threonine with methionine at codon 3192 of the DNAH1 protein (p.Thr3192Met). The threonine residue is moderately conserved and there is a moderate physicochemical difference between threonine and methionine. This variant has not been reported in the literature in individuals with DNAH1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
