Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001378477.3(NYX):c.590T>C (p.Leu197Pro), citing Invitae Variant Classification Sherloc (09022015): In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. This missense change has been observed in individual(s) with incomplete stationary night blindness, nyctalopia, exotropia, and nystagmus. Meets CRC. (Invitae). The frequency data for this variant in the population databases is considered unreliable, as metrics indicate insufficient coverage at this position in the ExAC database. This sequence change replaces leucine with proline at codon 202 of the NYX protein (p.Leu202Pro). The leucine residue is highly conserved and there is a moderate physicochemical difference between leucine and proline.
