Uncertain significance for Familial hemophagocytic lymphohistiocytosis 4 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003764.4(STX11):c.319A>G (p.Met107Val), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the STX11 gene (transcript NM_003764.4) at coding-DNA position 319, where A is replaced by G; at the protein level this means replaces methionine at residue 107 with valine — a missense variant. Submitter rationale: This sequence change replaces methionine with valine at codon 107 of the STX11 protein (p.Met107Val). The methionine residue is moderately conserved and there is a small physicochemical difference between methionine and valine. This variant is present in population databases (rs761875800, ExAC 0.002%). This variant has not been reported in the literature in individuals affected with STX11-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
