NM_001110556.2(FLNA):c.3844G>A (p.Ala1282Thr) was classified as Uncertain significance for Oto-palato-digital syndrome, type II; Heterotopia, periventricular, X-linked dominant; Frontometaphyseal dysplasia; Melnick-Needles syndrome by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals with FLNA-related conditions. This variant is present in population databases (rs782466840, ExAC 0.002%). This sequence change replaces alanine with threonine at codon 1282 of the FLNA protein (p.Ala1282Thr). The alanine residue is highly conserved and there is a small physicochemical difference between alanine and threonine. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
