Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001017995.3(SH3PXD2B):c.1067G>A (p.Arg356Gln), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces arginine with glutamine at codon 356 of the SH3PXD2B protein (p.Arg356Gln). The arginine residue is highly conserved and there is a small physicochemical difference between arginine and glutamine. This variant is present in population databases (rs145215054, ExAC 0.05%). This missense change has been observed in individual(s) with primary congenital glaucoma (PMID: 22509100). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
