Description

This variant causes an A to G nucleotide substitution at the +3 position of intron 50 of the COL3A1 gene. Splice prediction tools are inconclusive regarding the impact of this variant on RNA splicing. To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with COL3A1-related disorders in the literature. This variant has been identified in 6/251146 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.

Description

The c.4254+3A>G intronic variant results from an A to G substitution 3 nucleotides after coding exon 50 in the COL3A1 gene. This variant was identified in one or more individuals with features consistent with vascular Ehlers-Danlos syndrome (EDS) and segregated with disease in at least one family (external communication). This nucleotide position is not well conserved in available vertebrate species. In silico splice site analysis for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
