This sequence change replaces arginine, which is basic and polar, with leucine, which is neutral and non-polar, at codon 155 of the NKX2-6 protein (p.Arg155Leu). This variant is present in population databases (rs534202593, gnomAD 0.1%), and has an allele count higher than expected for a pathogenic variant. This variant has not been reported in the literature in individuals affected with NKX2-6-related conditions. ClinVar contains an entry for this variant (Variation ID: 570512). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt NKX2-6 protein function.
