Description

The p.E1401G variant (also known as c.4202A>G), located in coding exon 26 of the CFTR gene, results from an A to G substitution at nucleotide position 4202. The glutamic acid at codon 1401 is replaced by glycine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.

Description

This sequence change replaces glutamic acid with glycine at codon 1401 of the CFTR protein (p.Glu1401Gly). The glutamic acid residue is moderately conserved and there is a moderate physicochemical difference between glutamic acid and glycine. This variant is not present in population databases (ExAC no frequency). This missense change has been observed in individual(s) with congenital absence of vas deferens (CAVD) (PMID: 26277102). ClinVar contains an entry for this variant (Variation ID: 53921). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
