Description

This sequence change affects a donor splice site in intron 13. It is expected to disrupt mRNA splicing and likely results in an absent or disrupted protein product. While this particular variant has not been reported in the literature, truncating variants in BRCA2 are known to be pathogenic (PMID: 20104584). Experimental studies on a different variant affecting this nucleotide (c.7007+1G>C) have shown that this sequence change can result in two aberrant transcripts, both of which are predicted to encode truncated proteins (PMID: 21394826, 22505045), suggesting that this nucleotide is essential. For these reasons, this variant has been classified as Pathogenic.
