p.Arg69Cys (CGC>TGC): c.205 C>T in exon 3 of the BCS1L gene (NM_004328.4). The R69C variant has not been published as a mutation, nor has it been reported as a benign polymorphism to our knowledge. The R69C variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position that is conserved in mammals. In silico analysis predicts this variant is probably damaging to the protein structure/function. Missense mutations in nearby residues (R73C, S78G) have been reported as pathogenic mutations, supporting the functional importance of this region of the protein. Therefore, this variant is a strong candidate for a pathogenic mutation, however the possibility that it is a benign variant cannot be excluded. The variant is found in MITONUC-MITOP panel(s).
