Segmental restricted hindbrain gene expression. Vertebrate rhombomeres (r1–8) are analogous to the Drosophila segments and are shown in this schematic of the anterior neural tube and forebrain expansion. Below the neural tube is shown the expression pattern of individual genes (dark orange or black, strong expression; light orange or gray, weak expression; no bar, no expression). These genes include signaling molecules, such as fibroblast growth factor 3 (Fgf-3); transcription factors, such as kreisler and Krox-20; Eph family tyrosine kinase receptors, such as Sek and Ebk genes; and the Hox homeobox genes Hox-a and Hox-b. Note that each of the genes is expressed in a spatially restricted manner. These overlapping expression domains create unique combinations of transcription factors within each rhombomere which are considered, in turn, to regulate the phenotype of the cells in each hindbrain region.

It is not likely that signals from the retina will control scleral growth directly. Instead, retina-derived chemical messengers probably activate signalling cascades that create a chain reaction of signalling, which moves through the RPE, choroid and sclera (arrows). For example, dopamine and/or nitric oxide signalling in the retina can cause changes in light adaptation, which might then result in the release of retinoic acid from the choroid. Choroidal retinoic acid release could then result in inhibition of scleral proliferation, and inhibition of myopia. Although all the molecules listed in the diagram have been shown to affect eye growth, the places where they may act are still theoretical, and have not yet been proven conclusively. Integrative physiology through nuclear receptor (NR)–mediated crosstalk. Nutrient-sensing NRs amplify or propagate nutrient signals through local and long-range hormone actions. In the fed state, intestinal farnesoid X receptor (FXR) activation resulting from bile acid release produces fibroblast growth factor (FGF) 15/19. FGF15/19 limits hepatic bile acid synthesis and promotes glycogen and protein synthesis to store nutrients. Postprandial carbohydrate and lipid influx activate a hepatic peroxisome proliferator-activated receptor (PPAR) δ–controlled lipid synthesis program. The phosphocholine PC(18:0/18:1) is secreted as a long-range hormone to promote muscle fat utilization. Feeding activates adipose tissue PPARγ, which, in turn, controls the release of multiple adipokines. Locally, FGF21 amplifies PPARγ signaling and promotes further adipogenesis. FGF1 is another paracrine signal that is essential for healthy adipose tissue remodeling and for maintaining whole-body insulin sensitivity. Systemically, adiponectin enhances lipid oxidation in muscle and liver, whereas leptin increases satiation by acting on the central nervous system. During fasting, hepatic PPARα activation secretes FGF21, whose pleiotropic effects, including elevated adipose tissue lipolysis and hepatic ketogenesis, are important for adaptive fasting response. In addition, FGF21 interferes with the circadian clock in the brain and modulates physical activities
