Special sequences in the 3' untranslated region (UTR) of unstable mRNAs are responsible for their unusually rapid degradation. As indicated, AU-rich sequences found in the 3' UTR of many short-lived mRNAs cause a rapid removal of the poly-A tail, which in turn makes the RNA unstable. Other mRNAs contain sequences in their 3' UTR that serve as sites for specific endonucleolytic cleavage.

Three normal mRNAs with very different half-lives. The continuous rapid degradation of the mRNA molecules that encode various growth factors allows their concentration to be changed rapidly in response to extracellular signals. A signal for rapid degradation in the 3' untranslated region (UTR) determines the half-life of the growth factor RNA (see Figure9-85). Histones are needed mainly to form the new chromatin produced during DNA synthesis; a large change in the stability of their mRNAs helps to confine histone synthesis to the S phase of the cell cycle.

An RNA kissing loop complex in HIV. The figure shows the self-complementary kissing loop involving an untranslated sequence, DIS, that is thought to contribute to genomic RNA dimerization and/or packaging (Paillart et al. 1996; Berkhout and van Wamel 1996). The HIV-1 DIS sequence is highly conserved. (A) The two loops separated. Note that base pairs will not form between the six self-complementary bases at the tip of the loop. (B) “Kissing,” or base-pairing of the six bases at the tip of the loops; (C) the two loops base-paired to one another. Proteins (not shown) may help stabilize these complexes in vivo.
