Insertional mutagenesis. A, The process is independent of genes carried by the retrovirus. Retrovirus, for example, mouse mammary tumor virus (MMTV), infects a human cell. The proviral deoxyribonucleic acid (DNA) is integrated into the host genome in infected cells. Rarely, the provirus inserts near a protooncogene (eg, int-1) and activates the protosoncogene. In this example the process activates a gene called Wnt-1 (formerly called int-1) and produces breast cancer in mice infected with the mouse mammary tumor virus (MMTV). The sites of MMTV integration observed in 19 different tumor isolates are indicated by arrows. Note that the insertions can activate transcription of the Wnt-1 gene from distances of more than 10,000 nucleotide pairs away and from either side of the gene. This effect is attributed to a powerful enhancer DNA sequence present in the terminal repeats of the MMTV genome.
