Gap junction communication in the vascular wall. (A) Schematic representation of connexins assembling to form gap junctions is shown. Gap peptides such as Gap27, are connexin mimetics which possess conserved sequence homology with the second extracellular loop domains of the connexins and inhibit their activity possibly by disrupting their assembly. (B) Oligomerization of six connexins forms a connexon which may serve as a hemichannel and contribute to paracrine signaling by releasing for instance ATP. When a connexon dock with another connexon from the adjacent cell, they form gap junctions. (C) In the vascular wall, endothelial cells are connected to other endothelial cells (EC) and vascular smooth muscle cells to other vascular smooth muscle cells (VSMC) by homocellular gap junctions. Endothelial cells can also form heterocellular gap junctions with vascular smooth muscle cells. The incidence of heterocellular gap junctions increases when the size of the blood vessels decreases.
