Chemokines fall mainly into two related but distinct groups: the CC chemokines, which in humans are mostly encoded in one region of chromosome 4, have two adjacent cysteine residues in their amino-terminal region; CXC chemokines, the genes for which are mainly found in a cluster on chromosome 17, have an amino acid residue between the equivalent two cysteines. These chemokines can be divided further by the presence or absence of an amino acid triplet (ELR; glutamic acid–leucine–arginine) preceding the first of these invariant cysteines. All the chemokines that attract neutrophils have this motif, while most of the other CXC chemokines, including the chemokines reacting with CXCR3, 4, and 5 lack it. A C chemokine with only one cysteine at this location, and fractalkine, a CX₃C chemokine, are encoded elsewhere in the genome. Each chemokine interacts with one or more receptors, and affects one or more types of cell. A comprehensive list of chemokines and their receptors is given in Appendix III.

Blockade of chemokine activity by poxvirus-encoded vCKs an vCKBPs. Chemokines form a gradient by interacting with GAGs at the surface of endothelial cells and other surfaces, and binding of chemokines to chemokine receptors in leukocytes induce the expression of adhesion molecules and migration of leukocytes through the endothelium to areas of virus infection. Poxviruses have evolved three complementary mechanisms to block chemokine activity. First, the expression of the vCK MC148 by MCV that binds to chemokine receptors and prevents activation by host chemokines. Second, the soluble MV vCKBP-1, encoded by M-T7, that binds the GAG binding domain of a wide range of chemokines and may prevent the correct localization of chemokines at the surface of endothelium. Third, the secreted vCKBP-2 from MV (M-T1) and VV (35 kDa) binds chemokines with high affinity and blocks their interaction with cellular receptors.

Mechanism of chemokine inhibition by the CKBPs encoded by pathogens. Chemokines are presented to the leukocytes on the surface of endothelial cells where they interact with GAGs. The chemokine receptors expressed at the surface of leukocytes interact with chemokines and intracelular signalling is triggered to induce cell migration. The CKBPs sequester chemokines and inhibit their interaction with either chemokine receptors or GAGs. The secreted CKBPs may be specific for chemokines, such as the gammaherpesvirus M3 or the poxvirus 35-kDa protein, or may be part of a protein with dual cytokine specificity, interacting with chemokines and other cytokines, such as TNF (poxvirus CrmB) or IFN-γ (MYXV M-T7). The CKBP gG encoded by alphaherpesviruses is expressed as a membrane protein at the virion surface that is proteolitically cleaved to produce a secreted protein.

Modulation of the chemokine system by viral proteins. Chemokines (CKs) mediate their biological effect by interacting with specific seven-transmembrane-domain chemokine receptors (CKRs) at the surface of cells. Viruses encode chemokine homologs (vCKs) that function as agonists, binding to chemokine receptors and transducing signals, or as antagonists, occupying receptor binding sites and preventing the binding of host chemokines. Viruses also encode chemokine receptor homologs (vCKRs), expressed at the cell surface, and chemokine binding proteins (vCKBPs) that sequester chemokines in solution.
