Figure 11.14Attachment of actin filaments to adherens junctions

Cell-cell contacts at adherens junctions are mediated by cadherins, which serve as sites of attachment of actin bundles. In sheets of epithelial cells, these junctions form a continuous belt of actin filaments around each cell. The cadherins are transmembrane proteins that bind β-catenin to their cytoplasmic domains. β-catenin interacts with α-catenin, which serves as a link to actin filaments.

(A) Adherens junctions, in the form of adhesion belts, between epithelial cells in the small intestine. The beltlike junction encircles each of the interacting cells. Its most obvious feature is a contractile bundle of actin filaments running along the cytoplasmic surface of the junctional plasma membrane. (B) Some of the molecules that form an adherens junction. The actin filaments are joined from cell to cell by transmembrane adhesion proteins called cadherins. The cadherins form homodimers in the plasma membrane of each interacting cell. The extracellular domain of one cadherin dimer binds to the extracellular domain of an identical cadherin dimer on the adjacent cell. The intracellular tails of the cadherins bind to anchor proteins that tie them to actin filaments. These anchor proteins include α-catenin, β-catenin, γ-catenin (also called plakoglobin), α-actinin, and vinculin.

Homophilic interactions between cadherins mediate two types of stable cell-cell adhesions. In adherens junctions, the cadherins are linked to bundles of actin filaments via the catenins (see Figure 11.14). In desmosomes, desmoplakin links members of the cadherin superfamily (desmogleins and desmocollins) to intermediate filaments (see Figure 11.34).

At the early 8 cell stage, blastomeres are characterised by a intercellular adhesion mediated by E-cadherin. At the compacted 8 cell stage, in addition to the formation of additional adherens junctions, some tight junctions appear on the apical side. At the 32 cell stage, gap junctions appear in addition to the existing adherens and tight junctions.
