Chronology of the major AD pathological events. The pathological studies mainly in the 1980s established the chronology of the major pathological events at least in neocortex shown in the figure. In fact, the presence of senile plaques, NFTs, and degenerated neurons is necessary for the definitive diagnosis of AD. There has been a consensus that dysfunction or degeneration of neuronal synapses seems to exist between Αβ deposition or PHF formation and neurodegeneration.
