Biochemical activities of the GDI. The GDI serves as a multi-functional regulator of signalling by Rho GTPases. The three distinct biochemical activities of the GDI (1) inhibition of nucleotide dissociation, (2) inhibition of GTP hydrolysis, and (3) membrane release, are illustrated in relation to the GTP-binding and hydrolytic cycle of the Rho proteins.
