Non-LTR retrotransposition model. In certain poly(A) retrotransposons such as R2Bm and probably in others as well, priming of reverse transcription is mediated by a nick in the target site. Thus, in these elements, cutting of target site DNA precedes initiation of reverse transcription. The nick in R2Bm target DNA need not be in a sequence complementary to the 3′ end of R2Bm template RNA for priming to occur (Luan et al. 1993).
