These T-cell independent responses occur rapidly, with antibody appearing in 48 hours after infection, presumably because there is a high frequency of precursors of the responding lymphocytes so that little clonal expansion is required. In the absence of antigen-specific T-cell help, only IgM is made and, in mice, these responses therefore work mainly through the activation of complement, which is most efficient when the antibody is of the IgM isotype.

As shown on the outside of the circle, lymphocytes can be divided into T cells bearing T-cell receptors (detected with anti-CD3 antibodies), B cells bearing immunoglobulin receptors (detected with anti-immunoglobulin antibodies), and null cells including natural killer (NK) cells, that label with neither. Further divisions of the T-cell and B-cell populations are shown inside. Using anti-CD4 and anti-CD8 antibodies, α:β T cells can be subdivided into two populations, whereas γ:δ T cells are identified with antibodies against the γ:δ T-cell receptor and mainly lack CD4 and CD8. A minority population of B cells express CD5 on their surface (see Section 7-28).

Antigen-sampling and B cell-switching sites for induction of intestinal IgA responses. The classical inductive sites are constituted by GALT, which is equipped with antigen-sampling M cells, T cell areas (T), B cell follicles (B) and antigen-presenting cells (APCs). Class switching from IgM to IgA occurs in GALT and mesenteric lymph nodes (MLN); from here primed B and T cells home to the lamina propria (LP) via lymph and blood. Primed B cells may also migrate from solitary follicles directly into the LP. The IgA+ cells differentiate into plasma cells that produce dimeric IgA with J chain ((IgA/J+), which becomes secretory IgA (S-IgA). Bone marrow-derived IgM+ B2 cells (CD5-) may give rise to pentameric IgM (IgM/J+) and S-IgM. B1 cells (here indicated as CD5+) from the peritoneal cavity reach LP by an unknown route, perhaps via MLNs. These IgM+ cells may switch to IgA within the LP under the influence of APCs that have sampled luminal antigen as dendritic cells within the epithelium (EP).
