Description

During October 2010, we collected several hundred flowers at the stage of the big bud from tea cultivar Fudingdabaicha [Camellia sinensis (L.) O. Kuntze cv. Fudingdabaicha]. Those flowers were dissected into three parts: petals, pistils and stamens. Flower parts were frozen in liquid nitrogen immediately and then shipped on dry ice to BIG Inc., in Shenzhen, China.Total RNA was extracted using RNeasy Plant Mini Kit system (Qiagen) according to the manufacturer's protocol from three flower parts separately. Subsequently, RNA integrity was confirmed using the Agilent 2100 Bioanalyzer with a minimum integrity number value of 8. For cDNA preparation, beads with Oligo(dT) are used to isolate poly(A) mRNA after equal amounts of total RNA from three parts were pooled together.
