Description

A glycerol stock of SMOOTH colonies (Sample ID 66060, a child of Sample ID 184, BioSample SAMN03275447 ) was plated on blood agar base + hemin (BABH) medium and grown overnight at 37C under 5% CO2 to generate SMOOTH colonies. A heavy streak of these SMOOTH colonies was struck out again on BABH yielding consistently SMOOTH colonies on BABH. A single colony (source material Sample ID 209472) on the BABH plate was selected for aerobic culture in chemically defined minimal medium (CDMM) in a 50 ml conical with 15 mL CDMM for 21 hours; 191 rpm 37C. Caps lightly cracked open. DNA was extracted from this culture for PacBio HiFi sequencing.
