This BioProject presents the transcriptomic analysis of the deep-sea zoantharian Zibrowius primnoidus, collected at 255 m depth in the Sicily Channel, Italy, to characterize its toxin repertoire using a bulk RNA-seq approach. Twelve polyps were preserved in RNA later, washed in PBS, and processed using a TRIzol-based RNA extraction with magnetic beads. After purification (Mag-Bind Total Pure NGS kit), RNA integrity was assessed via TapeStation (High Sensitivity RNA Screen Tape Assay). Library preparation used the Zymo-Seq RiboFree Total RNA Kit for rRNA depletion, followed by paired-end 150 bp sequencing on an Illumina NovaSeq 6000, generating 90,741,564 raw reads. Primary bioinformatics included base calling, demultiplexing, and adapter masking with Illumina BCL Convert v3.9.31.
